We have recently reported that there is a strong association between GRM6 and lung cancer and that altered expression levels of GRM6 might contribute to the occurrence of lung cancer. In this study, we examined the genomic structure of the GRM6 gene, including structural variation, mutation, CpG methylation of the coding and flanking sequences, and potential regulation by cigarette smoke. The results indicated that the GRM6 gene may be located in the minimal region of 6p21.3-p22.1. The entire GRM6 gene, with the exception of exon 1, was found to be located on a 50 kb genomic region flanked by the sequences of the highly homologous genes IRX1 and SERPINA1. There were no mutations detected in any of the exons or the introns of the gene. However, the GRM6 gene was highly methylated in the coding regions, suggesting that methylation of the GRM6 gene may be involved in regulating its expression. CpG sequences in GRM6 were not methylated in either a lung cancer cell line (NCI-H441) or a normal lung cell line (IMR-90). Thus, the mechanism of GRM6 expression in normal lung cell lines is distinct from that in cancer cells, suggesting a different function for GRM6 in lung cancer cells. Finally, cigarette smoke condensate (CSC) exposure also induced methylation of GRM6 gene in lung epithelial cells (BEAS-2B).
